Yes, you can do this both statically and with the starter cranking the engine. 
Statically you should verify that the resistance varies between infinity and 
something less than 5 Ohms.

First thing to check is that when you turn the key ON, you hear a relay click 
and then ~1 second later you hear another click. This verifies that the brain 
has power and ground and that much of the wiring is okay. During the time 
between clicks the fuel pump should run.
